Alkaline phosphatase is elevated in various diseases and the separation of alkaline phosphatase isoenzymes is useful clinical laboratory method for detecting organs involved especially in patients with solitary elevation of alkaline phosphatase. Ager gel electrophoresis and physicochemical tests were done with sera 160 normal persons, 20 patients with hepatobillary diseases, 13 patients with F.U.O., 10 patients with bone diseases and 11 pregnant women. The physiochemical tests employed were heat inactivation and various inhibitors such as urea, EDTA and L-phenylalanine.
Results demonstrated 4 distinct isoenzyme bans in various diseases and only 2 bands in normal persons on electrophoresis. Alkaline phosphatase from patients with hepatobiliary diseases, F.U.O. and bone diseases were labileto urea and heat inactivation, whereas alkaline phosphatase from pregnant women was rather heat stable. The inhibition rates of alkaline phosphatase activity by heat and urea in children were greater than those of adults.
